    Wildfires Impact Assessment on PM Levels Using Generalized Additive Mixed Models

    Wildfires are relevant sources of PM emissions and can have an important impact on air pollution and human health. In this study, we examine the impact of wildfire PM emissions on the Piemonte (Italy) air quality regional monitoring network using a Generalized Additive Mixed Model. The model is implemented with daily PM10 and PM2.5 concentrations sampled for 8 consecutive years at each monitoring site as the response variable. Meteorological data retrieved from the ERA5 dataset and the observed burned area data stored in the Carabinieri Forest Service national database are used in the model as explanatory variables. Spline functions for predictive variables and smooths for multiple meteorological variables’ interactions improved the model performance and reduced uncertainty levels. The model estimates are in good agreement with the observed PM data: adjusted R2 range was 0.63–0.80. GAMMs showed rather satisfactory results in order to capture the wildfires contribution: some severe PM pollution episodes in the study area due to wildfire air emissions caused peak daily levels up to 87.3 µg/m3 at the Vercelli PM10 site (IT1533A) and up to 67.7 µg/m3 at the Settimo Torinese PM2.5 site (IT1130A)

    Volatile organic compound emissions in relation to plant carbon fixation and the terrestrial carbon budget

    A substantial amount of carbon is emitted by terrestrial vegetation as biogenic volatile organic compounds (VOC), which contributes to the oxidative capacity of the atmosphere, to particle production and to the carbon cycle. With regard to the carbon budget of the terrestrial biosphere, a release of these carbon compounds is regarded as a loss of photosynthetically fixed carbon. The significance of this loss for the regional and global carbon cycles is controversial. We estimate the amount of VOC carbon emitted in relation to the CO2 taken up, based on our own enclosure and micrometeorological flux measurements of VOC emissions and CO2 exchange within the Mediterranean area and the tropical rainforest in Amazonia and on literature data. While VOC flux estimates are small in relation to net primary productivity and gross primary productivity, the amount of carbon lost as VOC emissions can be highly significant relative to net ecosystem productivity. In fact, VOC losses are of the same order of magnitude as net biome productivity. Although we must assume that large amounts of these reemissions are recycled within the biosphere, a substantial part can be assumed to be lost into longer-lived oxidation products that are lost from the terrestrial biosphere by transport. However, our current knowledge does not allow a reliable estimation of this carbon loss

    Mammary gland-specific regulation of GNRH and GNRH-receptor gene expression is likely part of a local autoregulatory system in female vizcachas (Rodentia: Chinchillidae)

    In addition to key mammotrophic hormones such as the pituitary prolactin (PRL) and the ovarian steroids progesterone and estradiol, there are local factors that modulate the tissue dynamics of the mammary glands during pregnancy and lactation. By immunohistochemistry and RT-PCR, we found local transcription and translation of gonadotropin-releasing hormone (GNRH), GNRH receptor (GNRHR), PRL and PRL receptor (PRLR) in mammary glands of adult vizcachas during pregnancy and lactation. Both GNRH and GNRHR showed a lag between protein expression and gene transcription throughout the gestational period: while the highest transcription levels of these genes were recorded at early-pregnancy, the epithelial immunoexpressions of both showed their maximum during lactation. RIA results corroborated the presence of GNRH in mammary glands at all the analyzed stages and confirmed the maximum amount of this peptide in the lactating group. Significant amounts of GNRH were detected in milk samples as well. Conversely, PRL and PRLR shared similar protein and gene expression profiles, all exhibiting maximum values during lactation. GNRH peptide content in mammary glands of females with sulpiride-induced hyperprolactinemia (HP) was significantly lower than that of control females (CT). Although PRL mRNA levels remained unchanged, there was a marked increase in the α-lactalbumin (LALBA) transcription in mammary glands of HP- vs CT-females. These results suggest that after targeting mammary glands, PRL stimulates the expression of milk protein genes, but also, tempers the local expression of GNRH. Mammary gland-explants supplemented with a GNRH analogue (GN-explants) had no differences in terms of PRLR or LALBA transcription levels compared to CT-explants, so the mammary PRLR signaling would not appear to be modulated by GNRH. Yet, mRNA expression levels of both GNRH and the GNRHR-downstream factor, EGR1, were significantly higher in GN-explants compared to that of CT which would point to a GNRH-positive feedback mechanism. In summary, the local coupled expression of GNRH, GNRHR and EGR1 in the mammary gland throughout pregnancy of vizcachas, the PRL-dependent mammary GNRH secretion as well as the GNRH positive feedback on its own transcription suggest an autocrine-paracrine regulatory mechanism and propose an active role for GNRH in mammary gland tissue remodeling.Fil: Corso, María Clara.     Awareness, discussion and non-prescribed use of HIV pre-exposure prophylaxis among persons living with HIV/AIDS in Italy: a Nationwide, cross-sectional study among patients on antiretrovirals and their treating HIV physicians

    Background: Before Pre-Exposure Prophylaxis (PrEP) was officially recommended and made available, a few surveys among gay and bisexual men, and persons living with HIV/AIDS (PLWHA), identified an informal use of antiretrovirals (ARVs) for PrEP among HIV-negative individuals. Before PrEP availability in Italy, we aimed to assess whether PLWHA in Italy shared their ARVs with HIV-negative individuals, whether they knew people who were on PrEP, and describe the level of awareness and discussion on this preventive measure among them and people in their close circle. Methods: Two anonymous questionnaires investigating personal characteristics and PrEP awareness, knowledge, and experience were proposed to HIV specialists and their patients on ARVs in a one-week, cross-sectional survey (December 2013-January 2014). Among PLWHA, a Multivariable Logistic Regression analysis was conducted to identify factors associated with PrEP discussion with peers (close circle and/or HIV associations), and experience (use in close circle and/or personal ARV sharing). Results: Eighty-seven specialists in 31 representative Infectious Diseases departments administered the questionnaire to 1405 PLWHA. Among specialists, 98% reported awareness, 65% knew the dosage schedule, and 14% had previously suggested or prescribed PrEP. Among PLWHA, 45.6% were somehow aware, discussed or had direct or indirect experience of PrEP: 38% "had heard" of PrEP, 24% were aware of studies in HIV-negative individuals demonstrating a risk reduction through the use of ARVs, 22% had discussed PrEP, 12% with peers; 9% reported PrEP use in close circle and 1% personal ARV sharing. Factors predictive of either PrEP discussion with peers or experience differed between men and women, but across all genders were mainly related to having access to information, with HIV association membership being the strongest predictor. Conclusions: At a time and place where there were neither official information nor proposals or interventions to guide public policies on PrEP in Italy, a significant number of PLWHA were aware of it, and approximately 10% reported PrEP use in their close circle, although they rarely shared their ARVs with uninfected people for this purpose. Official policies and PrEP availability, along with implementation programs, could avoid risks from uncontrolled PrEP procurement and self-administration practices
